Live cell imaging is separated into three categories based on the type of product: equipment, consumables, and software. On the basis of technology, live cell imaging is further separated into Fluorescence Recovery after Photobleaching and High-Content Screening. Fluorescence recovery after photobleaching (FRAP) is a method for determining the kinetics of diffusion through tissue or cells. It may analyze the two-dimensional lateral diffusion of a molecularly thin film made up of fluorescently labeled probes or analyze individual cells. This approach is particularly useful in biological research on protein binding and cell membrane diffusion. Additionally, the surface deposition of a fluorescing phospholipid bilayer makes it possible to characterize hydrophilic (or hydrophobic) surfaces in terms of surface structure and free energy.
In biological research and drug development, a technique called high-content screening (HCS), also known as high-content analysis (HCA) or celomic, is used to find compounds such as small molecules, peptides, or RNAi that modify a cell’s phenotypic in a desirable way. High content screening, then, is a particular kind of phenotypic screen carried out in cells that involve the study of complete cells or components of cells with simultaneous readout of a number of parameters. High-throughput screening (HTS), in which thousands of chemicals are simultaneously examined for their activity in one or more biological assays, is similar to HCS; however, the outputs of HCS experiments are more complicated cellular phenotypes. The synthesis of cellular byproducts like proteins may rise or decrease as a result of phenotypic changes, and the cell’s morphology (outward appearance) may also alter. Therefore, automated microscopy and image analysis are common in HCA. The term “screening” distinguishes high-content screening from high-content analysis, which may be high in content but low in throughput. High-content screening implies a level of throughput unlike high-content analysis, which does not.
